The Effect of Freezing-thawing on Dna Extraction from Cryptosporidium Oocysts in Fecal Samples
نویسنده
چکیده
The DNA of Cryptosporidium Oocysts was extracted from fecal samples by using Commercial Wizard Genomic DNA purification general kit, tow freezingthawing protocols have been compared in this study: deep freezer for three cycles and liquid nitrogen for fifteen cycles, the results indicate that the second protocol was more influential in obtaining a sufficient amount of DNA from Oocysts of Cryptosporidium spp in fecal samples.

Enzyme-linked immunoassay for detection of Cryptosporidium antigens in fecal specimens.
Cryptosporidium sp. is a ubiquitous 4- to 6-micron protozoan parasite infecting the intestinal tract of humans. It causes mild to fulminant diarrhea in patients, especially immunocompromised persons, and it may be hard to detect by microscopic fecal examination. An indirect, double-antibody enzyme-linked immunosorbent assay (ELISA) was developed using specifically produced goat and rabbit antis...

Abstract Background: Usual diagnostic methods for detection of Cryptosporidium include smear preparation, modified Zeihl-Neelsen staining, microscopic examination and observation of oocysts. No studies have been done to compare PCR and Zeihl-Neelsen staining in Iran. This study was done to determine the positive predictive value, (PPV) and the negative predictive value (NPV) of PCR in compariso...
متن کاملInternal Amplification Control for a Cryptosporidium Diagnostic PCR: Construction and Clinical Evaluation
Various constituents in clinical specimens, particularly feces, can inhibit the PCR assay and lead to false-negative results. To ensure that negative results of a diagnostic PCR assay are true, it should be properly monitored by an inhibition control. In this study, a cloning vector harboring a modified target DNA sequence (≈375 bp) was constructed to be used as a competitive internal amplifica...
متن کاملProtocol for DNA extraction of Cryptosporidium spp. oocysts in fecal samples.
Molecular characterization of Cryptosporidium spp.oocysts in clinical samples is useful for public health since it allows the study of sources of contamination as well as the transmission in different geographical regions. Although widely used in developed countries, in Brazil it is restricted to academic studies, mostly using commercial kits for the extraction of genomic DNA, or in collaborati...
متن کاملOccurrence of Cryptosporidium oocysts in fecal samples submitted for routine microbiological examination.
During a 7-month period, 2,252 fecal samples submitted for routine microbiological examination from 1,621 patients were screened for Cryptosporidium oocysts by the auramine staining method with Kinyoun acid-fast stain as the confirmatory stain. Cryptosporidium oocysts were detected in fecal samples from 19 (1.2%) patients, 18 of whom had gastroenteritis. Diarrheic stools from 14 of these 18 pat...
